Work Flow: TWIST
Protocol: Initial QC TWIST v3
Step: Qubit, Quantit, and Aggregate

As well as:

Work Flow: TWIST
Protocol: Library Prep TWIST v2
Step: Aliquot samples for enzymatic fragmentation TWIST

Description:

We used to always speedvac samples but now we don't. I think that some calculations should therefore be adjusted so that it is easier for lab people.

Basically it would be really helpful for lab people to see the actual amount that samples could max have as an input based on the concentration, not the total amount or 250 ng. I would like a column in the quantit and qubit step as well as the aggregation step where we see the "Actual amount input to prep". This will basically only be important for samples where the concentration is less than 8.33 ng/ul and the volume more than 30 ul. For example a sample with 4 ng/ul may have a large volume of 100 ul and therefore lims now thinks the input will be 250 ng. It will actually only be 120, which is way less and we may have to contact the customer regarding this. We now have to calculate these values manually and this is tedious and can go wrong, so this would be helpful! @saschaconradt hope you agree!

In the aliqoutation step I want this to be the same too - we currently have to manually "fix" the values calculated by lims as it will always count on speedvaccing if the total amount is more than 250 ng. See here: https://clinical-lims.scilifelab.se/clarity/work-complete/477434. I want the Amount needed (ng) and the volumes of H2O and sample to be correctly calculated without the speedvaccing as an option!

Is this possible?

Perhaps this could also be done in wgs RC? @saschaconradt what do you think?

Yes that is a good idea.
