Hi everyone!
First of all, thanks to the Baker lab for this amazing tool. I wanted to ask the community about their experience on guiding the model towards the desired structure. For example, when I want the designed protein to have a certain shape, I just add a random helix or sheet at a location which will guide the design and tell the model to connect that helix to the designed backbone.
This approach sometimes makes the designs to look more like something I want, but I wanted something a bit less explicit. Have any of you achieve a reasonable backbone design using fold conditioning, or more interestingly, writing your own auxiliary potentials? If yes, can you please share your story on setting up those things (fold conditioning and potentials)?
