I'm using the Mayflash Wii Classic Controller Adapter For Pc (https://www.amazon.com/Mayflash-Wii-Classic-Controller-Adapter/dp/B0089O4EB0) with a Wii Nunchuk connected. When I test with https://gamepad-tester.com I can clearly see the values changing when I move the joystick around, so I know that the nunchuk is talking to the computer. However, the HID-Wiimote Control Center says "No Wii Remotes connected", and I don't see any options to try to interact with the connected nunchuk. Is this a supported use case?

When connecting Wiimotes, have Dolphin running at the same time (you do not need a game open), and press the red SYNC button on the back.
Also, if the Wiimote is the only controller you're using (with the nunchuk), plug the extension into the bottom of the Wiimote instead of a seperate adapter.
This issue sounds invalid. Looks like that Mayflash adapter presents fake DirectInput game controllers that most likely don't match any info for a Wiimote. You have bypassed the need to link a Wiimote to your PC to use the Nunchuk or potentially the Wii Classic Controller.
